Structural organization and functions of the cytoskeleton Eukaryotic cells are able to change their shape, move, move organelles, and separate chromosomes during cell division, thanks to a set of proteins that make up the cytoskeleton.

The cytoskeleton consists of 3 elements (structures): 1 microtubules 2 actin filaments (microfilaments) 3 intermediate filaments The chemical composition and location of these structures are different. They perform a number of important functions; including support.

Microtubules Long tubes, 2425 nm in diameter, whose walls consist of a helical protein called tubulin. Microtubules are thin, hollow organelles that grow from one end (plus the end) by the addition of tubulin molecules. The other end is anchored in the centrosome (minus end).

Centrosome - called the microtubule organizing center (MTOC), is localized in the center of the cell near the nucleus. Microtubules are dynamic structures that can grow, shorten, and also have a high rate of assembly and disassembly (for example, the mitotic spindle).

Functions of microtubules: participation in the very act of movement, since they are structural elements (this is intracellular transport, for example: movement of synaptic vesicles); participation in cell division; formation of a system of fibers along which vesicles and organelles move (movement of substances from the Golgi complex to the ER).

The connection between microtubules and the clinic Drug therapy for cancer patients involves the destruction of the mitotic spindle with substances that disrupt polymerization (colchicine, vinblastine). These drugs bind to microtubules and inhibit the formation of the mitotic spindle,

prevent the divergence of chromosomes and thus destroy rapidly dividing cancer cells. To treat breast cancer, the drug Taxol is used, which acts on the microtubule apparatus, stopping the rapid division of cells.

Microfilaments (actin filaments) Consist of the protein actin. Actin is found in many eukaryotic cells, accounting for up to 5% of the total cell protein. Most cells contain a dense network of actin filaments. These are thin protein filaments with a diameter of -5 -7 nm, forming the cellular cortex, giving mechanical strength to the surface of the cell, allowing it to change its shape.

Intermediate filaments Found in the cytoplasm of eukaryotic cells. These are tough, durable protein fibers, stretch-resistant polypeptides, that form a characteristic “basket” near the core, and from there they move along curved trajectories along the periphery. The thickness is 810 nm, i.e., intermediate between the diameters of microtubules and actin filaments.

The functions of intermediate filaments are to resist mechanical pressure in epithelial cells; they participate in the formation of the nuclear lamina. That is, in addition to the mechanical, protective, the formation of a transcellular network to counteract external loads.

Intermediate filaments form several proteins: Keratins - 20 different keratins are found in epithelial cells alone, 8 heavy keratins - specific for hair and nails; Vimentin and related proteins – distributed in fibroblasts and endothelial cells;

Intermediate filaments provide resistance to mechanical stress, but some mutations in keratin genes alter the organization of keratin filaments, which are found in the basal layer of the epithelium. Such genetic changes increase the skin's sensitivity to mechanical damage. Examples are

ordinary and bullous pemphigus - blisters appear on the skin (especially on the palms, soles, around the mouth, torso, neck, scars form, nail detachment).

Neurofilaments - the cytoskeleton in nerve cells is formed from 3 types of proteins: low molecular weight proteins (NF-Z) with low molecular weight; medium molecular weight neurofilament proteins (NFM); high molecular weight neurofilament proteins (NF-H).

Neurofilaments are located along the neuron, giving elasticity to the long processes of neurons (Fig. 7-2, Art. 148, Chapter 7) The function of intermediate filaments is to participate in the formation of cell contacts - desmosomes (which connect neighboring epithelial cells) and hemidesmosomes - which attach epithelial cells to membrane on which they are located.

The nuclear lamina - or fibrous lamina - is a network of intermediate filaments located on the inner surface of the nuclear membrane and associated with nuclear pores. Unlike other intermediate filaments, lamins are destroyed during mitosis by fragmentation of the nucleus and nuclear membrane and are restored at the end of mitosis.

MATERNAL GENES COORDINATED THE EVOLUTION OF A CHILD’S MITOCHONDRIA (FROM 05/28/2019)

Mitochondria have long been known for their independence: it is believed that they can reproduce and fight with each other without “consulting” with the rest of the cell. A new study has discovered the limit of mitochondrial independence: it turns out that most of the mutations that occur in their DNA are not inherited. Descendants receive only “allowed” mutations, which somehow managed to be “harmonized” with the nuclear genome.

In search of an explanation for this phenomenon, a large group of British scientists studied in detail the inheritance of mitochondrial DNA in more than 1,500 mother-child pairs.

Each human cell contains hundreds or even thousands of mitochondria. If they all contain the same DNA sequence, this is called homoplasmy, but if different variants of mitochondrial DNA can be found in a cell, this is called heteroplasmy. Among one and a half thousand mothers and their children, scientists found heteroplasmy in 45%, that is, in the cytoplasm of their cells, at least 1% of the mitochondria contained sequence variants that differed from the rest of the mitochondria in the cell. All these options can be divided into three groups: inherited (the same in mother and child), lost in a generation (only in the mother) or newly formed (appear only in the child). It turned out that there were significantly more of the former - inherited ones (almost 20%) than lost or new ones (about 6%).

The researchers then compared their list of sequence variants with the largest mitochondrial DNA databases. It turned out that the participants in the experiment had new, previously unknown variants that had not yet been included in these databases. However, their chance of being inherited by a child turned out to be almost five times lower than that of the known variants. It turns out that for some reason mothers “prefer” to pass on more common variants to their offspring. However, the logic here is most likely strictly the opposite: some options receive the greatest advantage over others and therefore survive better in the egg and are passed on to the child.

However, we now have the first evidence that not only the most viable, but also the most “corresponding” mitochondria to the nucleus receive an advantage. And this may be the answer to the question why, after a nucleus is transplanted into a new egg, healthy mitochondria often become mutants. Perhaps it is nuclear genes that provoke their uneven survival.

• Developing the ability to think independently is based on the implementation of such a methodological approach to the study of any biological phenomenon or process, which is built on the following principle: norm - possible violations - consequences of violations - possible correction. This scheme allows students to form the basics of clinical thinking, stimulates students to work independently, to search for information and the ability to use it.

The organization of the educational process at the Department of Biology is carried out in accordance with the requirements of the State Educational Standard of Specialties, on the basis of which, together with the educational and methodological management and taking into account standard educational programs (TUP), annual working educational programs (RUP) have been developed.

The accepted form of laboratory work is frontal, when all students simultaneously perform the same work. Individualization is achieved by working with different objects. In some classes, work is carried out with one object, but under different conditions, which also allows you to diversify the experiment and teaches students to be independent.

One of the technologies that has been introduced and is being actively improved at our department is modular learning technology. At its core, the modular system for organizing the educational process is currently the most effective methodological teaching system.

In order to systematically monitor the effectiveness of student learning, several types of control are used. Control is based on the principle of constant, duplicate control, when a topic mastered at a certain level is controlled several times.

• Current control - verification of students' preparedness for one laboratory lesson using an express test survey.
• Modular control - assessment of students' knowledge of three or four lessons that make up a training module.
• The test is an assessment of students' mastery of laboratory classes: 19 classes in the 1st and 16 classes in the 2nd semester.
• The exam is an assessment of the success of mastering all the material in the biology work program during training at the department.

History of the Department of Medical Biology and Genetics

In 1931, the Faculty of Medicine of Kazan University was transformed into the Medical Institute. In the same year, the Department of Biology was created at the newly opened institute.

Educational and methodological work

The department has theoretical status and carries out teaching activities in the disciplines: “Biology”, “Medical Genetics”, “Regenerative Medicine”, “Anthropology”, “General and Medical Genetics”, “Molecular Biology”, “Fundamentals of Medical Genetics”. The management of educational biological practice (practice in obtaining primary professional skills and abilities, including primary skills and skills in research activities) of 1st year students of the specialty “Medical Biochemistry” is carried out.

Employees of the department participate in the implementation of educational programs in the following specialties: “General Medicine”, “Pediatrics”, “Dentistry”, “Medical Biochemistry”, “Clinical Psychology”, “Social Work”.

Citywide competitions for students in medical biology have been organized and held on the basis of the Department of Biology of Kirov State Medical University (since 2015).

Employees of the department are members of the expert commission on biology of the Volga Region Open Olympiad for schoolchildren “The Future of Medicine”.

Scientific work

The main directions of research work of the department:

• Morphogenesis. Regeneration.
• Development of cell technology methods (from 2010 to 2013)
• Study of the cytoarchitecture of the lymphoid organs of the digestive system of mammals
• Study of anthropometric characteristics and factors of a healthy lifestyle of junior medical students

2 patents for invention were received (2017). Based on the results of scientific activities, more than 200 works have been published, including in the journals of the Higher Attestation Commission, RSCI and Scopus.

Organized and held: interregional scientific and practical conference “Cell pathology and methods of its correction” (from 2006 to 2015), Intra-university student scientific and practical conference “Topical issues of medical genetics” with the participation of geneticists of the Moscow State Budgetary Health Center “Kirov Regional Children's Clinical hospitals" (2015, 2016, 2017, 2018, 2019).

Since 1989 academic year A student scientific circle has been working at the department for years. Students take part in grant work, youth innovation forums, the U.M.N.I.K. competition, and All-Russian student scientific and practical conferences with international participation. Many works were awarded diplomas of I, II, III degrees. The results of scientific research by students participating in the SSS served as material for candidate dissertations.

Educational work

The main directions of educational work of the department:

• Research work of junior students as a method of professional self-determination of University students
• Prevention of alcoholism, drug addiction, hookah smoking, HIV infection and other socially dangerous phenomena among students

Every year, teachers of the department are curators of 1st and 2nd year student groups. Head of the department Koledaeva E.V. awarded the diploma “Curator of the Year - 2016”.

Historical reference

The Department of Medical Biology and Genetics was organized in 1987 as a Department of Biology with a histology course. The first head of the department was Doctor of Medical Sciences, Professor A.V. Molodyuk. After the histology course was allocated to an independent department in 1993, Doctor of Medical Sciences, Professor A. A. Kosykh was elected head of the Department of Medical Biology and Genetics. Since August 2014, the department has been renamed the Department of Biology and the head of the department is Candidate of Biological Sciences, Associate Professor Elena Vladimirovna Koledaeva.
